I have a cPanel shared account where Sendgrid had previously been configured with CNAME under Zone Editor but not used anymore. Should those settings be removed before setting up and using MXroute or isn’t there any conflict?

I can’t see a scenario where any existing CNAME record for that would conflict. It should be fine.
